Abstract

Evidences from in vivo and in vitro studies have demonstrated the role of estrogens in the development and preservation of the central nervous system. In addition to the well- known classical or genomic mechanisms of actions of estrogens, evidences accumulated in recent years have substantiated the existence of alternati\e mechanisms of actions for these hormones, which appear to be particularly relevant in the phenomenon of neuroprotection. Interestingly, these alternative actions are mediated by membrane estrogen receptors whose molecular identities are not well understood. In order to investigate the molecular features of these membrane located estrogen receptors, we have used several antibodies directed to different regions of the nuclear estrogen receptor (ER) to analyse by western blot the purified plasma membrane proteins of septum-derived SN56 and hippocampal-derived HT22 cell lines. Furthermore, taking into account that glycosylation may be a post-traductional modification accounting for the observed different estrogen receptors molecular weights, we have explored here the putative glycosylation of membrane ER isoforms. Our results demonstrate, in both cell lines, the existence of a high degree of structural homology bet- ween nuclear and membrane ER counterparts, as well as a clear O-glycosylation pattern in the 80 kDa membrane isoform.
